Scientists reported today in the journal Science that in cells where is not properly controlled by normal housekeeping genes, the protein can combine with another protein and start genetic changes related to cancer. The studies by scientists at Johns Flopkins University and in the University Hospital Utrecht, the Netherlands, offer an explanation of why patients with colon cancer usually have a mutated form of a gene called adenomatous polyposis coli, or APC. In its normal form, APC is known as a tumor suppressor. But a mutated form of APC is found in about 85 percent of all colon cancer tumors. Patients who inherit mutated APC genes can develop thousands of colon tumors. In the studies, the researchers beta-cateni- beta-caten- in show that the normal APC acts to When clean cells of the APC gene malfunctions, however, the amount of in cells increases. When there is excessive the laboratory beta-cateni- n. beta-caten- in beta-cateni- n, studies showed, it combines with another protein, called Tcf-Le- f. This combination of proteins disrupts the normal genes in the cell, a process that can lead to the development of a malignancy. Dr. Mark Peifer of the University of North Carolina at Chapel Hill said in Science that discovery of the role of in colon cancer may be the "smoking gun" that explains on a molecular level what causes normal colon cells to become cancerous and how a flawed APC gene is related to the disease. Another study in Science, by researchers at Onyx Pharmaceuticals in Richmond, Cal., and at the National Cancer Institute, suggests that may also play a role in another form of cancer. The researchers found abnormally high levels of beta- beta-caten- in beta-caten- in catenin in seven of 26 human melanoma cell cultures. They" also found that the APC gene was flawed or missing in two of ft the cell lines. ' , When APC was added to cell lines lacking the normal f orm of ';' this gene, it wiped out the exces. sive beta-cateni- The researchers concluded that excessive may play a role in the formation of melanoma cancers.
